Transposition of An Insertion Sequence
- The transposase coded for by the insertion sequence
- makes blunt ended cuts in the donor (IS) DNA
- and staggered, sticky end cuts in the target DNA
- The transposase then ligates the blunt ends of the IS to the 5' single stranded ends of the target DNA.
- DNA pol I extends the 3' cut ends and ligase seals the strands.
